Ultra-performance liquid chromatography (UPLC) is a instrumentation with high power in separation, little organic solvent consumption and analysis speed over traditional HPLC.In this study, a rapid UPLC method was developed and used to quantify simultaneously neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid and hyperin in E.koreanum Nakai.Chromatography was performed on a Acquity BEH C18 column (50 mm ×2.1 mm, l.7μm) with the mobile phase of methanol and water containing 0.1% acetic acid; the whole run time was only 14 min.The correlation coefficient between concentration and chromatographic peak area of neochlorogenic acid, chlorogenicacid , cryptochlorogenic acid and hyperinwas over 0.9996 in the range of 0.5760~57.60、 1.806~180.6、 1.056~105.6 and 1.530~153.0 μg·mL-1, respectively.The contents of four components of 10 batches of samples collected from different product areas and markets were determined.The method is convenient and feasible for determination of the content of neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid and hyperin in E.koreanum Nakai.
