Corydalis yanhusuo is a well known traditional Chinese herbal medicine.It is widely used to promote blood circulation, reinforce vital energy and alleviate pain.The major constituents are protoberberine type alkaloids, which have also been proved to be responsible for its bioactivities.However, as strong bases, these compounds often suffer from sever peak tailing and even irrversable adsorption problems on silica based RPLC columns.To overcome the HPLC problems of such basic compounds, we developed a new kind of polar-modified RP stationary phases, with a so called "polar-copolymerized" preparation approach.One representative column, named C18HCE, exhibited excellent peak shape and high loadability for basic standards in our previous evalution, and it had been successfully applied for the efficient purification of alkaloids from Corydalis yanhusuo.However, due to the structure similarity of protoberberines, the selectivity of C 18HCE column was not good enough to separate all the constituents.
