The LIM homeodomain transcription factor Lmx lb has been suggested to be essential for the development of midbrain dopaminergic (mDA) neurons,mainly based on their loss in Lmxlb-/-mice.However,we report here that the role of Lmx1b in mDA neuron development is mediated via the isthmic organizer (IsO).We found that mDA neurons developed normally in tyrosine hydroxylase-cre,Lmx1bflox/flox or dopamine transportercre/-,Lmxlbflox/flox mice in which Lmx1b was selectively deleted in mDA neurons.Moreover,mDA neurons were recovered in Lmx 1b-/-mice when IsO activity was rescued by Wnt1-Lmx1b transgene at the mid/hindbrain boundary,indicating that the loss of mDA neurons in Lmx1b-/-mice was a consequence of the disruption of the IsO.In agreement with this observation,when inactivation of Lmx1b occurred after the formation of the IsO by Nestin-Cre,mDA neurons developed normally.Our results indicate that the IsO is essential for mDA neuron development,and Lmxlb expression in mDA neurons is dispensable for their development.
