In the early postnatal cerebellum,postmitotic granule cells migrate from the external granular layer (EGL) to the internal granular layer (IGL) along radial glial fibers.Although this migration event has been well described,the underlying cellular and molecular mechanisms remain largely unknown.We have developed a method of electroporation to reliably introduce DNA constructs into granule cell precursors in the superficial part of EGL in vivo and trace the migration and positioning of granule cells after interference with the expression of specific genes.Preliminary results showed that down-regulation of plexinA2 in cerebellar granule ceils disrupted theft positioning during development with defects in the radial migration and the neurite extension.
