The 43-kD growth-associated protein (GAP-43) is a unique membrane associated protein in the nervous system.It has attracted much attention because the properties of the protein are associated with axonal growth during development and synaptic plasticity,axonal regeneration in the adult.However,some reports have demonstrated that an increased expression of GAP-43 in vivo may induce cellular apoptosis during the development and regeneration in the nervous system,suggesting that the GAP-43 may play a dual role in different conditions.Based on this observation,a hypothesis that axonal growth can be realized towards attractive signals,and collapse of growth cone and then cell death in unfavorable condition following a brain injury,can be raised.In order to verify this hypothesis and explore the molecular mechanism underlying the involvement of GAP-43 into these processes,we obtained the GAP-43 gene from the rat,utilizing the PT-PCR and inserted into pGEM-T vector firstly,then amplified a series GAP-43 mutant in this system.
