Metabolic dysregulation have been considered to play an important role in cancer initiation and progression.The observed shift fi-om oxidative phosphorylation to increased glucose burning renders tumor cell susceptible to intoxification by reactive aldehydes such as methyl glyoxal (MGO) that is formed non-enzymatically from glycolytic intermediates.MGO is known for its mutagenic and antiproliferative impact as well as its ability to trigger cellular apoptosis.Protection of cells from this cytotoxic metabolite is conferred by glyoxalases (Glo) that convert MGO to non-toxic D-lactate using glutathion as co-subtrate.To prevent cell death tumor cells are capable to escape MGO intoxification by upregulation of Glo.This offers an elusive mean to kill tumor cells by inhibiting this enzyme.
