Effects of 3-week simulated microgravity and intermittent artificial gravity on apoptosis of abdomin

  This study was designed to investigate the effects of 3-week simulated microgravity and intermittent artificial gravity on apoptosis of abdominal aorta smooth muscle cells (AASMCs) in rats.Hindlimb unweighting rats (HU) were used to simulate the effects of microgravity, and standing countermeasure (STD, daily 1 h of-Gx gravitation) was used to simulate the effects of intermittent artificial gravity.Apoptosis of AASMCs was assessed by the means of the basis of TUNEL staining, and expressions of Bad, Fasl and Caspase-3 in abdominal aorta were observed by Western blot.Results showed that: (1) The percent of TUNEL staining positive cells of AAVSMCs increased significantly in HU rats and partially recovered in STD rats.(2) The expression of Caspase3, FasL and Bad were all increased significantly but that of Bcl-2 was decreased significantly in HU, and all changes partially recovered in STD.These findings suggest that the apoptosis of AASMCs may play an important role in the adaptive remodeling of vessels induced by simulated microgravity.
