METHODS: Culturing HT29 cells, in logarithmic growth phase, cells were treated with berberine (26.25, 52.5, 105, 210, 420μmol/L) and evodiamine(5, 10, 15, 20, 25, 30μmol/L) for 24, 48, 72, 96h, respectively, a drug dose-effect curve was made to search the optimum concentration and time, under which we tested the inhibition of berberine and evodiamine for HT29 cell proliferation by cell counting way and the telomerase activity By TRAP method.First, We diluted cells into single cell suspension, then adjusted its concentration to 5× 105/0.2mL, which was injected into nude mice left abdominal subcutaneous to produce tumor-bearing nude mouse model, then those mice were randomly divided into four groups: control group, into which injected saline; Zuojin Pill group, AZT(Azidothymidine)group and collaborative group.We observed the effects of Zuojin Pill and AZT on tumor growth and teiomerase activity.We also used 1,2-dimethyl hydrazine(DMH)to induce rat colorectal cancer model, rats were randomly divided into four groups, similarly, then killed after 11、 21、 34 weeks treatment in batches to observe the result of Zuojin Pill and AZT with rat colorectal cancer development.RESULTS: In vitro experiment, our results suggest the repressive function upon HT29 cell proliferation of evodiamine and berberine are positively correlated with their dosage.The suppression ratio of evodiamine (7.5,15,30 μmol/L) was 39.3% ± 2.13%, 52.8% ± 5.34% and 64.1% ± 7.19%(all p <0.01), while the percent of berberine (52.5,105,210μmol/L) was 44.1%± 3.97%, 55.9%± 4.12%, 65.3% ± 6.94%(all p <0.01), respectively.Berberine and evodiamine could effectively inhibit HT29 cell telomerase activity.In tumorbearing nude mice model, Zuojin Pill and AZT have no significant influence on tumor size and also have weak action upon telomerase activity.In DMH-induced rat colon cancer model, Zuojin Pill and AZT can obviously control the process of cancer in the early stage, the tumor incidence of control group、 Zuojin Pill group、 AZT group and collaborative group are respectively as follows: 20, 0, 10, 10 % in the first 11 weeks.But at later stage (34 weeks), there is no significant difference.CONCLUSION: In vitro trial, the major components of Zuojin pill can suppress telomerase activity and cell HT29 proliferation.In tumor-bearing nude mice model, Zuojin Pill and AZT have no clear effects on tumor size and have weak impact on telomerase activity.In DMH-produced rat model, Zuojin pill can repress the invasion and growth of the early rat carcinoma effectively, while in late cancer, this action is not obvious..
