After screening the β-galactosidase (lactase) gene of Bifidobacterium breve B24,the DNA library was constructed in pBR322 plasmid vector with BamHI,EcoRl,and HindlII and subcloned into pDrive vector for the sequencing.The PCR amplification fragments based on galA sequence was ligated into pET-36(B) over-expression vector,and the pET-36(B) plasmid containing a β-galactosidase gene was transformed into E.coli ER2566.One complete ORF encoded 691 amino acids (2076 bp) was the structural gene,LacA (galA) of at the f-galactosidase gene.The deduced protein had a 76,010 Dalton and a pI of 4.58.
