Isolation and Characterization of a Tonoplast Aquaporin Gene (L1TIP2) from Lily Leaves

  Aquaporins (AQPs) are membrane intrinsic proteins of the tonoplast and the plasma membrane that facilitate the transport of water and other small neutral molecules across these membranes.An aquaporin gene (L1TIP2, GenBank accession number: JN085950) was isolated from lily (Lilium Iongiflorum Thumb.) leaves by RT-PCR and RACE.The full cDNA sequence of L1TIP2 was 986 bp, containing 208 bp 3-UTR, 34 bp 5-UTR, and 744 bp open reading frame, which encoded 247 amino acids.Two highly conserved NPA (Asn-Pro-Ala) motifs, proposed as defining the specificity of the water channel can be found in the deduced protein.Phylogenetic analysis showed L1TIP2 was a member of the TIP2 (tonoplast intrinsic proteins) subfamily.Homology of nucleotide sequences between L1TIP2 and TIP2 of Gossypium hirsutum, Hordeum vulgare, Lolium perenne and Triticum aestivum was 77%, 74%, 73%, 73% and 72% respectively.Further research on the functional characterization of L 1TIP2 and its response to postharvest water stress is ongoing.
