AtlTPK2 is a member of inositol 1,3,4-trisphosphate 5/6 kinase family, which can phosphorylate inositol 1,3,4-trisphosphate to form inositol 1,3,4,5-tetrakisphosphate and inositol 1,3,4,6-tetrakisphosphate.The ITPK2 was constitutively expressed in Arabidopsis thaliana and a T-DNA knock-out mutant of AtlTPK2 was identified to study its physiological roles.Scanning electron microscopy analysis showed that the epidermis structure of seed coat was irregular in itpk2 seeds, resulting in the increased permeability of the seed coat to tetrazolium salts.Gas chromatography coupled with mass spectrometry of lipid polyester monomers in the cell wall confirmed a drastic decrease in composition of suberin and cutin which was associated with the permeability of the seed coat. These results indicate that AtlTPK2 plays a major and essential role in lipid polyester biosynthesis which builds lipophilic barriers of seed coat to protect the embryo from biotic and abiotic stresses.
