In developing novel drugs for the treatment of metabolic disorders, there has been an increasing recognition of the importance of multi-faceted action.Accordingly, the design of two or more therapeutic mechanisms into a single molecular entity is becoming an exciting frontier in therapeutics.Biologics are ideally suited to such modular design, as both chemical and recombinant strategies have been applied to generate mixed-function modalities.Examples can be drawn from early stage efforts in protein design, especially antibodies, but peptides as a class are as, or more, suitable as a platform for multifunctional designer therapies.In this talk, a specific design platform, phybrids (peptide hybrids), and its application to metabolic disorders will be described.The potential for improving multiple pharmaceutical and biological attributes by engineering, rather than screening, opens the door to dramatic and unprecedented biologic performance enhancement.
