Objective: Finding a specific agent will be useful for monitoring allorejection in clinic.Macrophage migration inhibitory factor (MIF) was reported to be one of the major cytokines involved in the allorejection.In this study, we evaluated whether 131I-anti-MIF mAb could be an efficient imaging reporter for monitoring allorejection.Methods: We established the mice skin allo-transplantation models and analyzed the expression of MIF within grafts by RT-PCR and immunohistochemistry.The model mice were injected with 185kBq of 131I-anti-MIF mAb or 131I-IgG (control) respectively.At day1, 7, 14 and 21 after transplantation, the percentage of uptake by organs and per gram tissues (%ID/g) was calculated.The concentration of 131I-anti-MIF mAb and 131I-IgG in the grafts were evaluated by the calculated T/NT (target-to-non-target) ratios.Whole-body images were obtained with storage phosphor screen.Result: The results showed that up-regulation of MIF expression was found in allografts but not in isografts..There was no difference between 131I-anti-MIF mAb (T/NT ratio was 7.46) and 131I-IgG (T/NT ratio was 7.37) in early stage of allograft rejection (Day 1).With the development of the allograft rejection, the T/NT ratio of 131I-anti-MIF mAb increased continually, at day 7 it was 11.63, until the crest-time of rejection (day 14) the T/NT ratio was17.13.The T/NT ratio of 131I-IgG group decreased during the progression of rejection, from 7.37 to 1.66.The whole-body autoradiography images showed that both of the imaging agents showed a few uptakes in the grafts at dayl.Nevertheless, with the development of the allograft rejection, the 131I-anti-MIF mAb showed much more clear images than the 131I-IgG, which is in accordance with its high T/NT ratio.Conclusion: The higher T/NT ratio and its specificity targeting implied 131I-anti-MIF mAb may be a valid method for facilitating the development of protocols to monitor allorejection.
