SNARE protein dependent exocytosis in microglia lysosomes

  As the major immunological cell in CNS,microglia is well studied in the field of cytolines release,but the release mechanism in microglia is largely unknown.Lysosome is considered as a terminal degradative compartment of the endocytic pathway,into which transport is thought to be unidirectional.However,specialized secretory vesicles regulated by calcium share characteristics with lysosomes,suggesting that lysosome exocytosis might be a generalized process present in most cell types.Here we showed that lysosome in microglia exhibits calcium and SNARE protein dependent exocytosis.Incubation of cultured microglia with FM dye specifically labels lysosomes,as identified by immunostaining with lysosome specific marker lamp-1.Time lapse imaging with confocal microscopy showed that application of ATP,glutamate,or ionomycin induced calcium-dependent exocytosis in lysosomes as revealed by the destaining of FM dye-labeled clusters.Furthermore,treatment of cultures with tetanus toxin (TeNT) that cleaves vesicle protein VAMP2 significantly inhibits lysosome exocytosis.Our results indicate that SNARE protein-dependent lysosome exocytosis may play essential role in microglia functions.
