Objective: To investigate the anti-tumor characterization of oncolytic virus, whose E1A gene is controlled by a renal cell carcinoma-specific promoter (G250).Methods: The oncolytic virus G250-Ki67 has a transgene of Ki67-siRNA, and E1B55 was deleted in G250-ZD55-Ki67.The tumor-specific expression ofE 1A and Ki67 was demonstrated via Western blot and immunohistochemistry staining.The tumorspecific cytotoxicity was evaluated via crystal violet staining and cell viability assays.Apoptosis induction for tumor cells was detected using Annexin Ⅴ-PE/7-AAD.All of the adenovirus expressed E1A protein in 786-0 and OSRC cell lines but not in ACHN and HK-2 cell lines.
