Bacterial persistence is a phenomenon in which heterogeneous gene expression in an isogenic cell population gives rise to a subpopulation,called persisters,that can tolerate antibiotic treatment.Persister formation has been previously attributed to the dormancy state of the cell in response to stress.Here we show by in vivo fluorescent imaging,bacterial persisters under β-1actam antibiotic treatment have less cytoplasmic antibiotic accumulation due to enhanced efflux activities,snggesting that the capability to dynamically pump antibiotics out of the cell is pertinent to persistence.For the underlying molecular mechanism,transcriptome sequencing reveals that a number of multi-drug efflux associated genes show higher expression levels in persisters.Using a fluorescent membrane potential indicator,we further demonstrate that persister cells undergo more frequent and dramatic membrane potential changes,which may drive the enhanced effiux activities.At last,we show the combinational use of bacterial efflux inhibitors and antibiotics can effectively reduce the number of persisters,which provides a new therapeutic strategy for fighting drug tolerance.
