外周血中食管鳞癌相关miRNA与circRNA及其靶基因间相互作用的生物信息学分析

目的:对外周血中食管鳞癌相关的7种miRNAs进行circRNAs及其靶基因的预测,分析其与circRNAs及靶基因间的相互作用. 方法:根据前期进行系统综述和meta分析所获得的miRNAs,分别采用starBase及miRWALK对miR-18a、miR-21、mi-25、miR-31、miR-185、miR-223、miR-373的circRNAs和靶基因进行生物信息学预测,利用Cytoscape软件对这7种miRNAs和预测所得到的circRNAs及相应的靶基因进行网络分析,并进一步通过DAVID软件对预测的靶基因进行通路分析. 结果:由starBase对这7种miRNAs预测的circRNAs数量分别为33、15、34、23、24、19和43个.其中SP140_hsa_circ_001735和SP140_hsa_circ_001283与miR-21、miR-25和miR-373等3种miRNAs相互作用,ISY1_hsa-circ_001859与miR-18a、miR-21和miR-31等3种miRNAs相互作用,ZNF264_hsa_circ_001634与miR-25、miR-31和miR-373等3种miRNAs相互作用.在miRWALK中7-8个以上数据库预测到的靶基因中,RSBN1基因是miR-25、miR-31、miR-185和miR-373等4种miRNAs相对应的靶基因,PDZD2基因是miR-18a、miR-21、miR-25和miR-31等4种miRNAs相对应的靶基因.RASA1基因是在2种预测范围内与miR-21、miR-31和miR-223等3种miRNAs相对应的靶基因.在7种miRNAs所预测的靶基因相关KEGG信号通路中,7-8个数据库以上共有的信号通路为TGF-beta信号通路、Cell cycle信号通路、ErbB信号通路、PI3K-Akt信号通路、Pathways in cancer和Signaling pathways regulating pluripotency of stemcells. 结论:本研究对外周血中食管鳞癌相关的miRNAs及其相互作用的circRNAs、靶基因和信号通路进行了生物信息学预测和网络分析,有助于进一步研究miRNAs在食管鳞癌发生、发展过程中的调控作用和方式.
