Mitochondria are the important organelles that affect tumor initiation,growth,survival,and metastasis.The mitochondrial proteostasis was largely maintained by HSP60,which is one of the major classes of ATP dependent chaperones in mitochondria and is one of the most conserved proteins from bacteria to mammals.The purpose of the present study is to characterize HSP60-mediated protein interaction network and downstream signaling pathways underlying tumorigenesis and progression.Using quantitative proteomics,we identified that HSP60 bound to 1000 mitochondrial proteins associated with the respiratory complexes,mitochondrial ribosomes,protein transportation machinery,and protein degradation system.The binding of HSP60 with NADH dehydrogenases maintained the integrity of respiratory complexes and HSP60 silencing downregulated expressions of NADH dehydrogenases,leading to overproduction of reactive oxygen species (ROS).Using the biochemical analysis,we revealed that excessive ROS modified HSP60[1]and activated AMPK signaling pathway[2].
