Triptolide, which is separated from Tripterygium wilfordii Hook F, has a wide range of pharmacological activities.Han et al reported that CYP2E1, 1A2, 3A1 and 2C11 were induced by triptolide in rats in an in vivo experiment.Rat primary hepatocytes provide a valuable tool for the CYPs induction investigation since it is easier for gene manipulation compared to the in vivo model.To investigate whether the rat primary hepatocytes is an alternative for the study of the effects of triptolide on CYPs, rat primary hepatocytes were separated from male Wistar rats (180 ~ 200 g) and treated with vehicle or TP (50 and 100 nmol/L).After 48-hour treatment, total RNA and proteins was collected, qRT-PCR assay was employed for the detection of CYPs mRNA levels, and Western Blot assay was used for protein expression investigation.
