Triptolide, a major active ingredient of Tripterygium wilfordii Hook F., has been widely used for treatment of autoimmune disease in China. However, narrow therapeutic window and severe hepatotoxicity limit its clinical application. The recent studies indicated that metabolic pathways participated in detoxification of triptolide. In our previous study, we found that P-gp was involved in biliary excretion and detoxification of triptolide in sandwich-cultured rat hepatocytes, but the role of P-gp in triptolideinduced hepatotoxicity in vivo is not clear.
