Modularized biological parts are imperative for the engineering of genetic systems.Therefore, as an engineering process to eliminate the widespread interactions between natural biological components, refinement is an urgent, substantial work.However, it is progressed at a slow pace.Here we developed a production line to refine promoters and operators, the major players of transcriptional control in prokaryotes.The line realized the functional decoupling of promoter core regions and operators in transcriptional control.As a result, we were able to develop a non-free parameter model to direct the manufacturing of transcriptional devices.As a proof, we exhaustively tested the combination of 6 promoters and 4 operator sets to biologically implement transcriptional devices.Further, 82 random combinations of 52 promoters and 35 operators were also tested.The results showed that the gene expression control using these devices is in high precision with the model prediction.
