The cell cycle is characterized by periodic gene expression and post-translational modification of proteins,which are involved in phase-specific processes.Cancer cells characteristically lost cell cycle control.Actin-related proteins (Arps) are subunits of both ATP-dependent chromatin remodelling and histone acetylase complexes that have been implicated in orderly cell cycle progression.To define the molecular role of Arps as a function of cell cycle regulation,genes encoding Arps have been cloned in a synthetic vector that allows tandem affinity purification of Arps and their associated protein partners and complexes.Subsequently,mass spectrometry identifies proteins and their modifications.One isoform of Arps shows difference with cell cycle-staged cancerous and non-cancerous human cells while one isoform does not fluctuate with cell-cycle progression and the third isoform seems harmful for the cancer cell line.
