Radical SAM is a recently discovered superfamily of enzymes that use a [4Fe-4S] cluster for reductive cleavage of S-adenosylmethionine to generate the 5′-deoxyadenosyl radical.Radical SAM enzymes have been implicated in a variety of biochemical reactions and biological processes,including biosynthesis of vitamins,coenzymes,and antibiotics,as well as DNA repair and RNA modification.Thus far,only 4 structures of Radical SAM enzymes have been determined and the information concerning sequence-structure-function relationships in the entire superfamily is incomplete.
