目的:单纯依靠血清学分型,已不能准确的反应出复杂多变的淋球菌株群的流行病学特征。营养型,质粒类型和血清型及耐药类型结合起来可以克服单一分型法的不足,有助于鉴定不同地区的流行株,证实在分布和流行上的变化。方法:收集,分离流行株,于桂敏培养基上传代一次。分别接种于完整的GGM平板和不同的非完整GGM平板,置二氧化碳温箱培养,根据菌株在完整的GGM平板或缺少某一氨基酸或次黄嘌呤和尿嘧啶的平板上的生长情况以判定其营养型质粒DNA的抽提采用Kodo-Liu方法。质粒DNA与标准质粒一起进行琼脂糖凝胶电泳,根据DNA分子量的对数与迁移成反比的规律,制标准曲线,建立回归方程,计算出菌株质粒的分子量。结果:被检测的98株淋球菌流行株均能在完整的GGM平板上生长,部分菌株分别在缺少某一氨基酸或次黄嘌呤和尿嘧啶的非完整GGM平板生长不良。分型结果表明,该地区分离的菌株38.7%属于原养型(Wt);34%属于精氨酸、次黄嘌呤和鸟嘧啶营养缺陷型(AHU);17.35%属于脯氨酸缺陷型(PRO),余为其他营养型(Zie/Ser/PAU等)。全部98株淋球菌含有24.5Md、4.5Md和2.6Md三种质粒的占18.4%,含4.5Md和?
Objective: Relying solely on serological typing can not accurately reflect the epidemiological characteristics of a complex and variable gonococcal flora. Nutritional, plasmid type and serotypes and drug resistance types combined to overcome the deficiencies of the single typing method, help identify the epidemic strains in different regions, confirm the distribution and epidemic changes. Methods: The epidemic strains were collected and separated on the Gui-Min medium. Were inoculated on intact GGM plates and different non-intact GGM plates, incubated in a carbon dioxide incubator, and their nutrition was judged according to the growth of strains on intact GGM plates or on plates lacking a certain amino acid or hypoxanthine and uracil Plasmid DNA extraction using Kodo-Liu method.
