目的 :探讨CpG ODN对小鼠骨髓来源的树突状细胞 (bonemarrow deriveddendriticcells ,BMDC)分化成熟的影响。方法 :利用合成的含非甲基化CpG基序的寡核苷酸 (CpGmotifcontainingoligonucleotides,CpG ODN) ,通过酶切鉴定DNA制品CpG基序甲基化程度 ,FACS分析DC表型和内吞作用的变化 ,ELISA检测DC培养上清中IL 12 ,IL 6 ,IL 1β和TNF α和NO的含量。结果 :CpG ODN能够刺激DC前体细胞增殖 ,提高DCMHCⅡ类分子、B7 2和CD40的表达 ,明显增加IL 12等细胞因子的分泌 ,显著增强DC刺激初始型T细胞增殖。结论 :CpG ODN与LPS一样能够促进DC成熟进而增强抗原提呈功能
Objective: To investigate the effect of CpG ODN on differentiation and maturation of bone marrow derived dendritic cells (BMDC). Methods: CpG motif CpG motifs (CpG ODNs) were used to detect the degree of methylation of CpG motifs in DNA preparations. The changes of DC phenotypes and endocytosis were analyzed by FACS The contents of IL 12, IL 6, IL 1β, TNF α and NO in DC culture supernatants were detected by ELISA. Results: CpG ODN stimulated the proliferation of DC precursors, increased the expression of DCMHCⅡ, B7 2 and CD40, significantly increased the secretion of IL 12 and other cytokines, and significantly enhanced the proliferation of DC-stimulated naive T cells. Conclusion: CpG ODN, like LPS, can promote the maturation of DC and enhance the antigen presenting function
