目的观察加味大黄蟅虫颗粒对精索静脉曲张性不育模型大鼠的生精效应。方法按照随机数字表法将40只大鼠随机分为4组:假手术组、模型组、迈之灵组、加味大黄蟅虫颗粒组,每组10只。建立精索静脉曲张性不育大鼠模型,给药干预方法:假手术组、模型组每天给予生理盐水灌胃;迈之灵组大鼠每天给予迈之灵片54mg/kg;加味大黄蟅虫颗粒组每天给予加味大黄蟅虫颗粒(10g/kg)灌胃。4周后检测大鼠附睾精子的质量,检测大鼠睾丸组织生化酶学的活性,比较大鼠睾丸上皮显微超微结构的改变,免疫组化检测模型大鼠睾丸组织中Nrf2蛋白的表达。结果与假手术组比较,模型组大鼠附睾精子浓度低,睾丸病理改变明显,睾丸曲细精管层次紊乱,成熟精子数量减少或缺如;透射电镜下睾丸各级生精细胞坏死,线粒体空泡化,内质网扩张,细胞核染色质固缩、核崩解;免疫组化睾丸组织中Nrf2的棕黄色阳性表达的细胞辉度低。与模型组相比,迈之灵组和加味大黄蟅虫颗粒组精子浓度显著高于模型组,成熟精子数量和生精细胞形态结构明显恢复;免疫组化睾丸组织中Nrf2的棕黄色阳性表达的细胞辉度明显升高;迈之灵组和加味大黄蟅虫颗粒组超氧化物歧化酶(SOD)、α-葡萄糖苷酶的活性、果糖浓度与模型组比较有差异性(P<0.05)。结论加味大黄蟅虫颗粒能改善精索静脉曲张大鼠睾丸的病理损伤,提升睾丸组织中Nrf2的蛋白表达,对睾丸的精子发生发育具有一定的保护作用。
Objective To observe the effect of flavonoids from Rhubarb on the spermatogenesis in rats with varicocele infertility. Methods According to the random number table, 40 rats were randomly divided into 4 groups: sham operation group, model group, MaiZhiLing group and flavored rhubarb worm group, 10 rats in each group. To establish a varicocele-infertility rat model, administration intervention methods: sham-operation group, the model group were given saline daily; Maizhiling group rats were given daily matrine tablets 54mg / kg; flavored big yellow pinworm Granules group given flavored rhubarb worm particles (10g / kg) orally. After 4 weeks, the quality of epididymal spermatozoa was detected. The biochemical and enzymatic activities of testis in rats were detected. The ultrastructure of testis epithelium was compared. The expression of Nrf2 protein in testis tissue was detected by immunohistochemistry. Results Compared with the sham-operation group, the spermatozoa of the model group had low spermatozoa, obvious pathological changes of the testis, abnormal testicular seminiferous tubules and reduced or absent mature spermatozoa. The spermatogenic cells necrosis, Vacuolization, endoplasmic reticulum dilation, nuclear chromatin condensation and nucleus disintegration. Immunocytochemical staining of Nrf2 in the testis showed low brightness. Compared with the model group, the sperm concentration of maizarin group and flavobacteria was significantly higher than that of the model group, and the number of mature sperm and morphology of spermatogenic cells were restored obviously. The positive expression of Nrf2 in testis (P <0.05). The activity of superoxide dismutase (SOD) and α-glucosidase in the Mai Ling group and the flavonoids group were significantly higher than those in the model group (P <0.05). Conclusions Modified Rhizoma Imperatae can improve pathological damage of testis in rats with varicocele and enhance the expression of Nrf2 protein in testis tissue, which may have some protective effect on testicular spermatogenesis.
