目的研究由15个精氨酸组成的小肽硫酸R15对未分级肝素的拮抗作用。方法通过抗凝血因子Ⅱ试验对比硫酸鱼精蛋白和硫酸R15体外中和未分级肝素的能力;在肝素化的Wistar大鼠中评价硫酸R15体内中和情况。结果抗凝血因子Ⅱ试验结果表明,体外试验中硫酸R15有更强的肝素中和能力;在体内试验中硫酸R15和硫酸鱼精蛋白中和未分级肝素的能力相似,无统计学差异。结论硫酸R15与硫酸鱼精蛋白相比有良好的药效,是一种潜在的新型肝素拮抗剂。
Objective To study the antagonism of unfractionated heparin by R15, a small peptide of 15 arginine. Methods The anti-coagulation factor II test was used to compare the ability of protamine sulfate and R15 to neutralize unfractionated heparin in vitro. The neutralization of R15 in vivo was evaluated in heparinized Wistar rats. Results The results of anticoagulant factor Ⅱ test showed that in vitro experiments, R15 had stronger ability of neutralizing heparin. There was no statistical difference in the neutralization of unfractionated heparin between R15 and protamine sulfate in in vivo experiments. Conclusion Sulfuric acid R15 has good efficacy compared with protamine sulfate and is a potential novel heparin antagonist.
