目的比较加味四物汤不同提取工艺制备得到的样品(A、B、C、D)对小鼠镇痛作用和偏头痛保护作用,为进一步研究其作用机制提供前期药理学依据。方法采用小鼠醋酸扭体法记录小鼠在15 min内扭体次数,计算药物对扭体反应的抑制率。断头张口呼吸实验记录小鼠断头后张口喘息的时间。采用利血平致小鼠偏头痛模型,观察药物对血清超氧化物歧化酶(SOD)、谷胱甘肽(GSH)的活力和丙二醛(MDA)、钙离子(Ca~(2+))以及脑组织5-羟色胺(5-HT)含量的影响。结果加味四物汤样品A~D能明显延长小鼠断头张口喘息时间(P<0.05或P<0.01);A和C能明显提高小鼠扭体反应抑制率(P<0.05或P<0.01);能有效增强偏头痛模型小鼠血清SOD和GSH的活力(P<0.05或P<0.01),降低MDA和Ca~(2+)含量(P<0.05或P<0.01),提高脑组织的5-HT含量(P<0.05或P<0.01)。结论加味四物汤样品A和C有明显镇痛作用和偏头痛保护作用,即当归和川芎的挥发油以及处方的乙醇提取部分可能为其有效作用的物质基础,其作用机制可能与调节血液自由基和Ca~(2+)水平反常,以及脑组织的神经递质有关。

Conclusion Jiawei Siwu Decoction sample A and C have obvious analgesic effect and migraine protective effect, that is, the volatile oil of Angelica and Chuanxiong and prescription ethanol extract may be the material basis for its effective function, which may be related to the regulation of blood free radicals And Ca ~ (2+) levels abnormalities, as well as brain neurotransmitter.
