利用离体孵育脑薄片和放射免疫测定孵育液内精氨酸升压素(AVP)的方法,探讨了皮质酮(B)及牛血清白蛋白偶联皮质酮(B-BSA)对大鼠下丘脑薄片(含室旁核和视上核)释放AVP的快速影响。结果:(1)B和B-BSA在20min内均可明显地抑制AVP的释放,在10-7~10(-4)mol/L范围内均呈剂量-效应关系;但在每一剂量,B-BSA的快速抑制效应均弱于B;(2)RU486对B和B-BSA的快速抑制效应都有部分的阻断作用;但阻断B-BSA效应的最小剂量比阻断B者高一个数量级;(3)孵育液Ca~(2+)浓度升高,B和B-BSA的快速抑制效应均明显增强;孵育液中无Ca~(2+)则B和B-BSA的快速抑制效应有所减弱;但B-BSA效应的变化幅度不及B大。结果表明:(1)B与牛血清白蛋白共价偶联后,其快速抑制大鼠下丘脑薄片释放AVP的效应有所减弱;(2)B的快速抑制效应是由非基因组机制介导的,其作用可能是影响Ca~(2+)跨细胞膜流动的结果。
