目的分析类风湿关节炎(RA)患者外周血和关节滑液中Th极化细胞(Th1、Th17细胞)的表达。方法流式细胞技术测定RA患者外周血、关节滑液中CD4T细胞上IFN-γ+IL-17-(Th1)、IL-17+IFN-γ-(Th17)的表达,并与健康人比较,分析疾病活动指数(DAS28)和Th极化细胞表达量、Th1/Th17比值之间的关系。结果RA患者关节滑液Th1、Th17细胞的平均百分比与RA外周血、健康人外周血比较,差异有统计学意义(P<0.01)。RA患者关节滑液中Th1细胞、Th1/Th17细胞比值与DAS28指数呈正相关(P<0.01)。结论在RA疾病部位关节液中是Th1细胞占优势,且Th1细胞的表达与疾病活动性密切相关。
Objective To analyze the expression of Th-polarized cells (Th1 and Th17 cells) in peripheral blood and synovial fluid of patients with rheumatoid arthritis (RA). Methods The expression of IFN-γ + IL-17- (Th1) and IL-17 + IFN-γ- (Th17) on CD4 T cells in peripheral blood and synovial fluid of patients with RA were measured by flow cytometry. Analysis of disease activity index (DAS28) and Th polarization of cell expression, Th1 / Th17 ratio between the relationship. Results The average percentage of Th1 and Th17 cells in synovial fluid of RA patients was significantly higher than that of RA peripheral blood and healthy volunteers (P <0.01). The ratio of Th1 cells and Th1 / Th17 cells in synovial fluid of RA patients was positively correlated with DAS28 index (P <0.01). Conclusions Th1 cells are predominant in synovial fluid of RA disease sites, and the expression of Th1 cells is closely related to disease activity.
