目的:探讨即刻早期反应基因-1(immediate early response gene X-1,IEX-1)转染人卵巢癌细胞SKOV3后,对顺铂药物敏感性的影响。方法:提取并鉴定重组质粒pEGFP-N1-IEX-1,采用脂质体介导将重组质粒转染到人卵巢癌细胞SKOV3,RT-PCR检测转染前后IEX-1mRNA和蛋白的表达情况,采用四甲基偶氮唑蓝(MTT)法检测转染后卵巢癌细胞SKOV3增殖情况以及对顺铂敏感性的变化。结果:成功转染IEX-1基因后可检测到IEX-1mRNA的高表达;转染组细胞的增殖速度增快(P<0.05),但对顺铂的敏感性明显增强(P<0.05)。结论:IEX-1可增强卵巢癌细胞的体外增殖能力,并增强卵巢癌细胞体外对顺铂化疗的敏感性,为肿瘤的有效干预和治疗提供新的靶点。
Objective: To investigate the effect of immediate early response gene X-1 (IEX-1) on cisplatin-induced drug sensitivity in human ovarian cancer cell line SKOV3. Methods: The recombinant plasmid pEGFP-N1-IEX-1 was extracted and identified. The recombinant plasmid was transfected into human ovarian cancer cell line SKOV3 by lipofectamine. The expression of IEX-1 mRNA and protein before and after transfection was detected by RT-PCR. The MTT assay was used to detect the proliferation of ovarian cancer cell line SKOV3 and the sensitivity to cisplatin. Results: The high expression of IEX-1 mRNA was detected after transfection of IEX-1 gene. The proliferation rate of transfected cells was increased (P <0.05), but the sensitivity to cisplatin was significantly enhanced (P <0.05). Conclusion: IEX-1 can enhance the proliferation of ovarian cancer cells in vitro and enhance the sensitivity of ovarian cancer cells to cisplatin chemotherapy in vitro, and provide a new target for the effective intervention and treatment of cancer.
