为了研究中枢神经系统是否表达促血小板生成素 (TPO)的受体c mpl和TPO对神经细胞生长和保护作用 ,用免疫组织化学和RT PCR方法分析c mpl表达 ,用MTT ,annexinⅤ方法和流式细胞术检测、观察TPO对神经细胞生长和抗凋亡作用。结果表明 ,人中枢神经系统和小鼠神经细胞均表达TPO受体c mpl,人大脑半球和小脑及小鼠神经干细胞株C17.2在mRNA水平表达c mpl,进一步在蛋白水平证实TPO受体c mpl在人大脑半球神经细胞表达 ,TPO对C17.2细胞有生长促进作用和抗凋亡作用。结论 :本研究首次证实中枢神经系统表达TPO受体c mpl和TPO有促进体外神经细胞生长和抗凋亡的作用。
To investigate whether central nervous system (CNS) express cmpl and TPO, a receptor for thrombopoietin (TPO), on neuronal growth and protection, c mpl expression was analyzed by immunohistochemistry and RT PCR. MTT, annexin Ⅴ and flow cytometry Cytometry was performed to observe the effect of TPO on the growth and anti-apoptosis of nerve cells. The results showed that both human central nervous system and mouse neurons express TPO receptor c mpl, human cerebral hemispheres and cerebellum and mouse neural stem cell line C17.2 express c mpl at the mRNA level, further confirming that the TPO receptor c mpl is expressed in human cerebral hemispheres neurons, TPO has a growth promoting effect on C17.2 cells and anti-apoptotic effects. CONCLUSIONS: This study, for the first time, demonstrates that the expression of the TPO receptors c mpl and TPO in the central nervous system promotes the growth and anti-apoptotic effect of neural cells in vitro.
