应用倒置相差显微镜、扫描电镜、透射电镜和免疫细胞化学技术,对原代培养的神经细胞进行形态学观察。结果显示:接种后10h,少数细胞开始贴壁;24h时,大部分细胞贴壁,突起延长并互相连接,部分细胞NSE免疫染色呈阳性;3d时,胞体增大,突起互相连接成神经网络,胞核大,形态规则,胞质内含较多的粗面内质网和线粒体,高尔基复合体发达;7d时神经细胞晕光明显,胞体饱满,NSE免疫阳性细胞增多;14d时,神经细胞数目减少,有些神经细胞开始退化;21d时,神经细胞数目明显减少,突起萎缩,出现碎解。
The primary cultured neurons were observed morphologically by inverted phase contrast microscope, scanning electron microscopy, transmission electron microscopy and immunocytochemistry. The results showed that a few cells tended to adhere at 10h after inoculation. Most of the cells adherent, the protuberances were extended and connected to each other at 24h, some NSE immunostaining cells were positive. At 3d, the cell bodies were enlarged and the neurites were connected to each other, Large nucleus and regular morphology, more rough endoplasmic reticulum and mitochondria were contained in the cytoplasm, and the Golgi complex developed. On the 7th day, the cornea of halo cells was obvious, the cell body was full, and the number of NSE immunopositive cells increased. On the 14th day, Decreased, some neurons began to degenerate; 21d, the number of nerve cells decreased significantly, the process of atrophy, there collapse.
