目的:探讨脑源性神经营养因子(BDNF)在不同发育时期人胚胎脊髓中的表达变化及意义。方法:采用免疫组织化学和免疫印迹法对3周~8月人胚脊髓中BDNF蛋白进行定位、定量研究。结果:所检测的15个时段中,神经管上皮和室管膜上皮均可见BDNF阳性细胞,且随胚龄增加呈规律性变化;免疫印迹法显示6~9周,BDNF蛋白含量随胚龄逐渐增加,9周时达到高峰,3月后下降,6月后又有所增加但相对平稳。结论:BDNF在人胚胎脊髓发育特别是胚期脊髓的发育中发挥重要作用,并可能参与神经上皮神经干细胞的分裂增殖。
Objective: To investigate the changes and significance of brain derived neurotrophic factor (BDNF) in human fetal spinal cord at different developmental stages. Methods: The BDNF protein in human embryo spinal cord from 3 weeks to 8 months was localized and quantified by immunohistochemistry and Western blotting. Results: BDNF positive cells were observed in the neural tube epithelium and ependymal epithelium in 15 periods, which showed regular changes with the increase of embryonic age. Immunoblotting showed that BDNF protein content gradually increased with embryonic age from 6 to 9 weeks , Peaked at 9 weeks, declined after March and increased again after June but remained relatively stable. CONCLUSION: BDNF plays an important role in the development of spinal cord in human fetal spinal cord, especially in embryonic stage, and may be involved in the division and proliferation of neural stem cells.
