应用体外集落与液体培养、细胞形态学观察、DNA片段凝胶电泳及 DNA片段百分率测定等方法观察灵芝复方对人骨髓 CFU- GM生长及 HL- 6 0细胞增殖和凋亡的作用。发现灵芝复方在一定浓度范围内对人骨髓 CFU - GM生长有促进作用 ,而对白血病细胞系 HL - 6 0细胞集落生长则表现出剂量依赖性抑制 ;细胞形态学结果表明 ,灵芝复方对 HL- 6 0细胞凋亡具有剂量和时间依赖性诱导作用。 8和 16 mg/ m L 灵芝复方作用48h,可使 HL - 6 0细胞 DNA片段百分率明显升高 ;DNA片段凝胶电泳显示有明显的细胞凋亡“梯形”表现。提示灵芝复方可通过选择性诱导白血病细胞凋亡来发挥抗白血病作用
The effects of Ganoderma lucidum compound on the growth of human bone marrow CFU-GM and the proliferation and apoptosis of HL-60 cells were observed by in vitro colony and liquid culture, cell morphology observation, DNA fragment gel electrophoresis and DNA fragment percentage determination. It was found that Ganoderma lucidum compound promotes the growth of human bone marrow CFU-GM in a certain concentration range, but exhibits a dose-dependent inhibition on the colony growth of leukemic cell line HL-60 cells; the cell morphology results show that Ganoderma lucidum compound on HL- Apoptosis of 60 cells has a dose- and time-dependent induction. 8 and 16 mg/m L of Ganoderma lucidum compound for 48 h, the percentage of DNA fragments in HL-60 cells was significantly increased; the DNA fragment gel electrophoresis showed a clear “trapezoidal” appearance of apoptosis. It is suggested that Ganoderma lucidum compound can exert anti-leukemia effect by selectively inducing apoptosis of leukemia cells.
