目的 探讨缺氧对大鼠主动脉平滑肌细胞增殖的效应。方法应用免疫组织化学染色、3 H-TdR掺入、流式细胞仪检测技 术探讨缺氧对大鼠主动脉平滑肌细胞(ASMC)增殖的影响。结果缺氧 12 h其增殖细胞核抗原(PCNA)阳性反应颗粒增加,24 h明显增 加,其含量是常氧组 2.1倍。低氧 8 h可使 ASMC的3 H-TdR摄取量减少(P<0. 05),12 h则促进其摄取,24 h则明显较常氧对照组增多 (P<0.05)。流式细胞仪检测显示其G2/M期细胞比例明显增多(P<0.01),G0/G1期细胞比例明显减少(P<0.01),细胞内总蛋白含量 有所降低(P<0.01)、结论缺氧早期抑制或损害ASMC增殖,而后促使PSMC的表型改变,出现增殖。从一侧面证实缺氧可以直接促进ASMC增生。
Objective To investigate the effect of hypoxia on the proliferation of rat aortic smooth muscle cells. Methods The effects of hypoxia on the proliferation of aortic smooth muscle cells (ASMCs) in rats were investigated by immunohistochemical staining, 3 H-TdR incorporation and flow cytometry. Results After 12 hours of hypoxia, PCNA positive reaction particles increased, and the content of PCNA was increased by 2.1 times in 24 hours. Hypoxia for 8 h reduced the 3 H-TdR uptake of ASMC (P <0.05), up-regulated the uptake at 12 h and increased significantly at 24 h (P <0.05). Flow cytometry showed that the proportion of cells in G2 / M phase increased significantly (P <0.01), the proportion of cells in G0 / G1 phase decreased significantly (P <0.01), the content of total protein in cells decreased (P < 0.01) .Conclusions Hypoxia can inhibit or impair the proliferation of ASMC early, and then induce the phenotype of PSMC to proliferate. Confirmed hypoxia from one side can directly promote ASMC proliferation.
