目的:研究补骨方对大鼠去势后骨质疏松症的作用机制。方法:实验动物为普通级Wister大鼠,采用去卵巢方法复制绝经后骨质疏松症模型,将大鼠随机分为假手术组、模型空白组、雌二醇组、补骨方中药组、中成药仙灵骨葆胶囊(黔卫药准字10019),贵州仙灵药业有限公司生产,批号为000842组。同时,应用补骨方15周进行防治,用仙灵骨葆胶囊组、尼尔雌醇组作为阳性对照组,以假手术组和模型空白组作为实验对照组。采用放免方法检测骨钙素(BGP)、雌激素(E2)、促黄体生成素(LH)、促卵泡激素(FSH);对各组实验动物分别取胫骨、股骨用双能X线骨密度仪检测骨密度(BMD)。结果:①实验动物血生化指标比较:与模型空白组比较,补骨方组能明显降低BGP、的水平(P<0.05),提高E2的水平(P<0.05),降低LH、FSH激素水平。③实验动物离体股骨及胫骨骨密度比较:与模型空白组比较,补骨方组骨密度有显著性差异(P<0.05)。结论:补骨方可有效防治绝经后骨质疏松症。
Objective: To study the mechanism of Bu Gu Fang on osteoporosis after ovariectomy in rats. METHODS: The experimental animals were ordinary Wister rats. The postmenopausal osteoporosis model was replicated by ovariectomy. The rats were randomly divided into sham operation group, model blank group, estradiol group, Bugufang Chinese medicine group, and Chinese herbal medicine group. Pharmacology Xianling Gushao Capsule (Guangzi Yaozi 10019), produced by Guizhou Xianling Pharmaceutical Co., Ltd., and the batch number is 000842 group. At the same time, the application of Bu Gu Fang was controlled for 15 weeks. The Xianlinggu capsule group and Nilestriol group were used as positive control group and the sham operation group and model blank group were used as experimental control group. Radioimmunoassay was used to detect osteocalcin (BGP), estrogen (E2), luteinizing hormone (LH), and follicle-stimulating hormone (FSH); dual-energy X-ray absorptiometry was performed on the tibia and femur for each group of experimental animals. Bone mineral density (BMD) was measured. Results: 1 Comparison of blood biochemical indexes in experimental animals: Compared with the model blank group, Bugufang group can significantly reduce the level of BGP (P<0.05), increase the level of E2 (P<0.05), and reduce the levels of LH and FSH hormones. 3Comparison of bone density of isolated femur and tibia in experimental animals:Compared with the model blank group, there was significant difference in bone mineral density between Bugufang group (P<0.05). Conclusion: Bugu recipe can effectively prevent postmenopausal osteoporosis.
