目的 以 m R N A 差异显示技术对比研究具有不同转移潜能的癌系,克隆与肿瘤转移相关的基因。方法 以肺巨细胞癌细胞系 P G 具有不同转移潜能的亚系( B E1, L H7)为研究对象,应用 m R N A 差异显示技术克隆差异片段,经 Northern 杂交验证后测序,进入 Gen Bank 数据库检验同源性。结果 m R N A 差异显示技术对比高低转移癌系m R N A,分别建立了高低转移相关基因资源库。其中一个849 bp 片段在高转移亚系( B E1)低表达,而在不转移亚系( L H7)高表达,两者差异显著,经 Gen Bank Blastn 检索,与 G3 B P( Ras G T Pase Activating protein S H3 dom ain bindingprotein)同源性高达 99% 。结论 在高低转移癌系中 G3 B P的表达具有显著差异,提示 G3 B P与肿瘤的转移表型有一定相关性,为 G3 B P功能的研究提供了新的线索。
Objective To compare and study the cancer lines with different metastatic potential by m R N A differential display technology and clone the genes related to tumor metastasis. METHODS: The subclones (B E1, L H7) with different metastatic potentials of the giant cell lung cancer cell line P G were used as research objects. The differential fragments were cloned using the m R N A differential display technique and verified by Northern hybridization and sequenced to enter the Gen Bank. The database checks for homology. Results The m R N A differential display technique contrasted m R N A with high- and low-metastatic cancer lines, and established high and low metastasis-related gene pools. One of the 849 bp fragments was lowly expressed in the high metastatic subgroup (B E1) and highly expressed in the non-metastatic subgroup (L H7). There was a significant difference between the two, with Gen Bank Blastn searching and G3 B P (RasG T). Pase-Activating protein S H3 dom ain binding protein) has a homology of up to 99%. Conclusion There is a significant difference in the expression of G3 B P in high and low metastatic cancer lines, suggesting that G3 B P has a certain correlation with the metastasis phenotype of tumors, providing new clues for the study of G3 B P function.
