目的:建立HPLC同时测定粉葛饮片中葛根素、大豆苷元-8-C-芹菜糖-(1→6)葡萄糖苷和大豆苷3种黄酮类成分含量的方法。方法:采用Diamonsil C18色谱柱(4.6 mm×250 mm,5μm),以甲醇-0.02%磷酸(25∶75)为流动相,检测波长250 nm。结果:精密度、稳定性和重复性合格,3个成分的线性范围分别为0.123~0.737,0.024 9~0.150,0.021 7~0.130μg,3个成分的平均回收率分别为99.4%,99.0%,99.1%,测定了4批粉葛饮片中3个成分的含量。结论:该方法简便、快速、准确度高,可为粉葛饮片的质量控制提供参考依据。
OBJECTIVE: To establish a method for the simultaneous determination of three flavonoid constituents of Puerarin, Daidzein-8-C-Celery Sugar- (1 → 6) glucoside and daidzin in Powder Pueraria Tablets by HPLC. Methods: Diamonsil C18 column (4.6 mm × 250 mm, 5 μm) was used with mobile phase of methanol-0.02% phosphoric acid (25:75) as the mobile phase. The detection wavelength was 250 nm. Results: The precision, stability and repeatability were qualified. The linear ranges of the three components were 0.123 ~ 0.737, 0.024 9 ~ 0.150 and 0.021 7 ~ 0.130μg, respectively. The average recoveries of the three components were 99.4% and 99.0% 99.1%. The contents of three components in four batches of powder were determined. Conclusion: The method is simple, rapid and accurate. It can provide a reference for the quality control of Powder Pueraria lobata.
