目的:研究四磨汤对慢传输结肠5-HT信号系统兴奋性受体5-HT3、4的影响,探索理气中药改善结肠动力的作用机制。方法:选取SD大鼠,采用大黄灌胃法制造结肠慢传输型便秘模型,造模成功后,将36只大鼠再随机分为3组,一组为对照组灌蒸馏水,另两组为四磨汤干预高剂量组和低剂量组。中药干预组灌服四磨汤,连续灌服2周后处死,测炭末推进率,取出标本。Realtime-PCR检测5-HT3、4受体mRNA的表达,免疫组化检测石蜡切片5-HT3、4受体蛋白表达,计算机辅助图像分析表达情况。结果:四磨汤干预高剂量组及低剂量组大鼠炭末推进率高于对照组(P<0.05);两组大鼠结肠中5-HT3、5-HT4受体mRNA表达的ct值均高于对照组(P<0.05),高剂量组与低剂量组之间mRNA表达差异无统计学意义(P>0.05);四磨汤干预高剂量组及低剂量组大鼠结肠中5-HT3、5-HT4受体蛋白的灰度值均高于对照组(P<0.05),高剂量组与低剂量组受体蛋白的表达差异无统计学意义(P>0.05)。结论:四磨汤能够增加慢传输结肠兴奋性受体5-HT3、4的表达量,提示理气中药可能通过上调兴奋性受体5-HT3、4的表达来改善结肠动力。
Objective: To investigate the effect of Si mill decoction on the 5-HT3,4 excitatory receptor of 5-HT signaling system in slow transit and to explore the mechanism of Qi-qi traditional Chinese medicine in improving colonic motility. Methods: SD rats were selected and the slow transit constipation model was established by Rhubarb gavage. After successful modeling, 36 rats were randomly divided into 3 groups. One group was distilled water and the other group was 4 Mill soup intervention high-dose group and low-dose group. Traditional Chinese medicine intervention group, four mill soup, continuous feeding 2 weeks after sacrifice, measuring the rate of carbon powder, remove the specimen. Realtime-PCR was used to detect the mRNA expression of 5-HT3,4 receptor. The expression of 5-HT3,4 receptor protein in paraffin sections was detected by immunohistochemistry. Results: The effect of Sihuang Tang intervention in high-dose group and low-dose group was higher than that of the control group (P <0.05). The ct values of 5-HT3 and 5-HT4 receptor mRNA expression in the colon of the two groups were (P <0.05). There was no significant difference in mRNA expression between the high-dose group and the low-dose group (P> 0.05). The effect of Sihuang decoction on high-dose group and low-dose group of 5-HT3 , While the gray value of 5-HT4 receptor protein was higher than that of the control group (P <0.05). There was no significant difference in the expression of receptor protein between the high dose group and the low dose group (P> 0.05). CONCLUSION: Si mill decoction can increase the expression of 5-HT3,4, a slow-release colonic excitatory receptor, suggesting that Qi-regulating traditional Chinese medicine can improve colonic motility by up-regulating the expression of excitatory receptors 5-HT3,4.
