细胞毒素α-Sarcin 可切断核糖体大rRNA 链3′端的某个部位,使核糖体失活,这一作用机制正在分子水平加以阐明。α-Sarcin(简称 AS)是从一种巨曲霉培养液分离精制而得的抗肉瘤成分,是分子量为17,500的碱性蛋白质,是强细胞毒素。AS对肿瘤细胞增殖的抑制作用特别强,人们猜想这是因为与正常细胞比较,它更容易透过肿瘤细胞膜。通过对无细胞系统的研究,了解到毒素的靶是真核细胞的核糖体,但是 AS使60S 核糖体失活的机制,则完全不得而知。
Cytotoxicity of α-Sarcin can cut off a part of ribosomal large rRNA chain 3 ’end, so that the ribosome inactivation, this mechanism of action is being elucidated at the molecular level. α-Sarcin (AS) is an anti-sarcoma component isolated and purified from a culture of Aspergillus oryzae. It is a basic protein with a molecular weight of 17,500 and is a strong cytotoxin. AS has a particularly strong inhibitory effect on the proliferation of tumor cells, which is suspected because it is more easily transmitted through the tumor cell membrane than normal cells. By studying cell-free systems, it is known that toxins target the ribosomes of eukaryotes, but the mechanism by which AS inactivates 60S ribosomes is completely unknown.
