目的:探讨mmu-miR-141在早孕小鼠胚胎着床前、后子宫内膜组织中的表达及作用。方法:荧光定量PCR(FQ-PCR)及原位杂交检测早孕小鼠胚胎着床前、后子宫内膜mmu-miR-141的表达;MTT和流式细胞术检测孕第4日(d 4)子宫内膜基质细胞被mmu-miR-141抑制剂作用72 h后其细胞活力和细胞凋亡情况。结果:mmu-miR-141在小鼠胚胎着床后(d 6)子宫内膜组织中的表达明显低于着床前(d 4)(P<0.05),且表达于基质细胞;其表达下调能使基质细胞增殖活力下降(P<0.05),细胞凋亡增加(P<0.01)。结论:mmu-miR-141通过调控子宫内膜基质细胞的增殖或凋亡,在早孕小鼠胚胎着床前、后的子宫内膜组织中发挥重要的作用。
Objective: To investigate the expression and role of mmu-miR-141 in endometrial tissue before and after embryo implantation in early pregnancy. Methods: The expression of mmu-miR-141 in endometrium of preimplantation embryos was detected by FQ-PCR and in situ hybridization. MTT and flow cytometry were used to detect the expression of mmu-miR- Endometrial stromal cells were exposed to mmu-miR-141 inhibitor for 72 h after the cell viability and apoptosis. Results: The expression of mmu-miR-141 in endometrium was significantly lower than that before implantation (d 4) (P <0.05) and was expressed in stromal cells after embryo implantation (d 6) Can make the stromal cell proliferation activity decreased (P <0.05), apoptosis increased (P <0.01). Conclusion: mmu-miR-141 plays an important role in the regulation of endometrial stromal cell proliferation or apoptosis in endometrial tissues before and after embryo implantation in early pregnancy.
