目的 研究骨髓间充质干细胞定向分化为腱细胞的可能性。方法 通过电穿孔法将外源基因BMP12导入骨髓间充质干细胞中 ,诱导骨髓间充质干细胞定向分化为腱细胞 ;在形态学和分子生物学水平上对诱导后的细胞加以鉴定。结果 光镜下 ,诱导后的细胞形态发生明显改变 ;RT PCR结果表明 ,诱导后的细胞有BMP12和CollagenⅠ的mRNA表达 ,而没有CollagenⅢ的mRNA表达 ;诱导后98 39%的细胞呈CD4 4 + 、HLA DR-。结论 BMP12能够诱导骨髓间充质干细胞定向分化为腱细胞 ,间充质干细胞有可能成为肌腱组织工程种子细胞来源之一。
Objective To study the possibility of bone marrow mesenchymal stem cells differentiating into tendon cells. Methods The exogenous gene BMP12 was introduced into bone marrow mesenchymal stem cells by electroporation and induced to differentiate into bone marrow mesenchymal stem cells. The induced cells were identified on morphology and molecular biology. Results The morphological changes of the cells were observed under light microscope. RT-PCR results showed that BMP12 and CollagenⅠ mRNA expressions were detected in the induced cells without mRNA expression of Collagen Ⅲ, while 98 39% of the induced cells showed CD4 4 + HLA DR-. Conclusion BMP12 can induce bone marrow mesenchymal stem cells to differentiate into tendon cells. Mesenchymal stem cells may become one of the sources of tendon tissue engineering seed cells.
