目的:通过参灵草口服液腺苷检验方法优化,提高质量标准,减少对高效液相色谱仪及色谱柱的损害。方法:参灵草口服液在上机前采用有机试剂三氯甲烷或石油醚萃取除去参灵草口服液中的脂溶性干扰成分;试验不同比例流动相,比较参灵草口服液中腺苷的含量差异。结果:样品前处理方法采用石油醚或三氯甲烷萃取洗涤,目的在于除去脂溶性干扰物质,结果发现色谱图没有变化,此方法不可行。采用甲醇—水梯度法洗脱,结果色谱图腺苷主峰出峰快、分离良好、峰形尖锐、杂质峰无干扰。同时该流动相不会污染色谱柱和管路,能够提高色谱柱和高效液相色谱仪的使用寿命。结论:该方法适用于参灵草口服液腺苷含量测定,主峰峰形尖锐、分离良好,又可提高仪器与色谱柱的使用寿命。
OBJECTIVE: To improve the quality standards and reduce the damage to high performance liquid chromatography and chromatography column by optimizing the method of oral ginseng test. Method: Shenlingcao oral liquid was extracted with organic solvent chloroform or petroleum ether to remove lipid-soluble interfering components in Shenlingcao oral liquid before the machine was put on. The purpose was to remove the fat-soluble interfering substances and found no change in the chromatogram. This method is not feasible. Using methanol - water gradient elution, the results of the main peak of adenosine peak fast, good separation, sharp peak shape, impurity peak without interference. At the same time, the mobile phase does not contaminate the column and tubing, increasing the useful life of your column and HPLC. Conclusion: The method is suitable for the determination of adenosine in Shenlingcao oral solution. The peak shape of the main peak is sharp and the separation is good. The method can also improve the service life of instruments and columns.
