目的研究阿司匹林对糖尿病大鼠脂肪细胞肿瘤坏死因子-α(TNF-α)mR-NA表达的影响,为临床防治2型糖尿病及微血管并发症提供理论基础和实验依据。方法用高糖高脂饮食及腹腔注射小剂量链脲佐菌素方法建立2型糖尿病大鼠模型,分为模型组、阿司匹林组和西药对照组,另选正常大鼠作为正常组。阿司匹林组给予阿司匹林450 mg/(kg.d)灌胃,西药对照组给予二甲双胍134 mg/(kg.d)灌胃。给药1个月后检测血糖、血脂、血清胰岛素含量和脂肪细胞TNF-αmRNA表达的变化。结果模型组大鼠血糖、血脂、血清胰岛素含量明显上升,脂肪细胞TNF-αmRNA表达显著升高;阿司匹林组和西药对照组以上指标均显著降低,组间比较,阿司匹林组明显低于模型组和西药对照组(P均<0.05)。结论阿司匹林能有效降低2型糖尿病大鼠脂肪细胞TNF-αmRNA表达和血糖、血脂、血清胰岛素浓度,其作用机制可能是通过抑制炎症因子释放而改善了胰岛素抵抗状态,因此有可能成为治疗糖尿病并发症的新的有效药物。
Objective To study the effect of aspirin on the expression of TNF-αmR-NA in diabetic rats and to provide theoretical and experimental evidences for the prevention and treatment of type 2 diabetes mellitus and microvascular complications. Methods A rat model of type 2 diabetes mellitus was established by high glucose and high fat diet and intraperitoneal injection of low dose streptozotocin. The rats were divided into model group, aspirin group and western medicine control group, and normal rats were selected as normal group. Aspirin group given aspirin 450 mg / (kg.d) gavage, western medicine control group was given metformin 134 mg / (kg.d) gavage. Blood glucose, blood lipids, serum insulin levels and the changes of TNF-αmRNA expression in adipocytes were measured 1 month after administration. Results The levels of blood glucose, blood lipids and serum insulin in model group were significantly increased, and the expression of TNF-αmRNA in adipocytes was significantly increased. The above indexes in aspirin group and western medicine group were significantly lower than those in model group and western medicine Control group (all P <0.05). Conclusion Aspirin can effectively reduce the expression of TNF-αmRNA and the blood glucose, blood lipid and serum insulin in adipocytes of type 2 diabetic rats. Its mechanism may be that it can improve insulin resistance by inhibiting the release of inflammatory cytokines and thus may be a potential therapy for diabetic complications The new effective drug.
