本文采用HPLC分析RU 486及其代谢产物。分析用200mm×3.5mm ODS/YWG柱(青岛海洋化工厂担体),流动相为甲醇:乙腈:水42:28:30(V:V:V),pH4.7,流速1ml/min,紫外检测为302nm。在此色谱条件下,能使RU 486及其三种代谢产物得到很好分离。四种化合物的标准曲线在100~2800ng/ml浓度范围内,线性的相关系数为0.999。RU 486和其代谢产物去单甲基衍生物(RU 42633),丙炔醇衍生物(RU 42698)不同浓度的回收率均达90%左右,另一代谢产物去双甲基衍生物(RU 42848)的回收率为60%左右,本方法已用于RU 486及其代谢产物的临床药代动力学研究。
In this paper, HPLC analysis of RU 486 and its metabolites.
