为阐明胚胎巨噬细胞(macrophage,MΦ)的分化和卵黄囊内胚层(yolksacendoderm,YS-E)之间的关系,用近交系DA大鼠胚胎的卵黄囊内胚层细胞体外培养,观察了YS-E细胞的膜抗原表型、贴壁能力及吞噬功能的变化;用免疫组织化学方法观察了YS-E样细胞在鼠胚内的发生和分布,证实了从胚胎第10.5d开始,即卵黄囊和胚体之间的血液循环相通之后,在鼠胚体内的疏松结缔组织、肝原基和脑等处,可以观察到和MΦ有相同抗原表型的YS-E样细胞。以上结果提示,大鼠胚胎巨噬细胞的内胚层起源的可能性。
To clarify the relationship between differentiation of embryonic macrophage (MΦ) and yolksacendoderm (YS-E), yolk sac endoderm cells of inbred DA rats were cultured in vitro and YS -E cell membrane antigen phenotype, adherent ability and phagocytic function; observed by immunohistochemistry YS-E-like cells in the mouse embryo and distribution, confirmed from the embryo 10.5d beginning, that is, YS-E-like cells with the same antigenic phenotype as MΦ can be observed after the blood circulation between the yolk sac and the embryoid body is connected with loose connective tissue, hepatic primordium and brain in the embryoid body. The above results suggest the possibility of endoderm origin in rat embryonic macrophages.
