目的:比较胰腺癌患者与健康志愿者血清蛋白的差异以寻找候选血清肿瘤标志物,为胰腺癌早期诊断提供依据。方法:通过免疫亲合柱去除丰度最高的14种血清蛋白,用双向电泳分离蛋白并比较差异。选取在胰腺癌患者血清中明显差异表达蛋白点,行质谱鉴定和生物信息学分析。结果:建立了稳定性、重复性良好的凝胶蛋白图谱。对筛选出的在胰腺癌患者血清中明显差异表达的78个蛋白点,共有54个蛋白点被成功鉴定,其中胰腺癌患者血清中高表达者29个,低表达者25个。结论:胰腺癌患者、健康志愿者血清蛋白之间存在明显差异,通过蛋白质组学方法可筛选并鉴定出这些差异蛋白,后者有望为早期诊断胰腺癌提供依据。
OBJECTIVE: To compare the difference of serum proteins between pancreatic cancer patients and healthy volunteers in order to find candidate tumor markers for early diagnosis of pancreatic cancer. METHODS: Fourteen serum proteins with the highest abundance were removed by immunoaffinity column, and proteins were separated by two-dimensional electrophoresis and compared. We selected the differentially expressed protein spots in the serum of patients with pancreatic cancer, and identified them by mass spectrometry and bioinformatics analysis. Results: The stability and reproducibility of the gel protein profile was established. A total of 54 protein spots were successfully identified in 78 protein spots that were significantly differentially expressed in sera from patients with pancreatic cancer. Among them, 29 were highly expressed in serum and 25 were low in pancreatic cancer patients. Conclusion: Serum proteins of pancreatic cancer patients and healthy volunteers are significantly different. These differential proteins can be screened and identified by proteomics. The latter is expected to provide a basis for the early diagnosis of pancreatic cancer.
