目的研究中药贯众(DCN)提取物对体外培养的人肝癌细胞的抑癌活性。方法 DCN经粉碎、浸提、超速离心、减压浓缩、冷冻干燥得DCN提取物。人肝癌SMMC-7721细胞经DCN提取物处理不同时间后以MTT法测其对人肝癌细胞线粒体代谢活性的影响,以细胞计数法观察了其对细胞增殖的影响。结果(1)DCN提取物能明显抑制人肝癌细胞的生长,降低线粒体代谢活性;(2)经DCN提取物处理24h后的人肝癌细胞,既使更换为正常培养基,其线粒体代谢活性及细胞增殖与对照相比仍差异显著。结论中药DCN提取物含抑癌活性物质,能抑制体外培养的肝癌细胞增殖和降低线粒体代谢活性。
Objective To study the anti-cancer activity of traditional Chinese medicine (DCN) extracts on human hepatoma cells cultured in vitro. Methods DCN was crushed, extracted, ultracentrifuged, concentrated under reduced pressure and freeze-dried to obtain DCN extract. Human hepatocarcinoma SMMC-7721 cells were treated with DCN extract for different periods of time, and their effects on the mitochondrial metabolic activity of human hepatocellular carcinoma cells were measured by MTT assay. Cell proliferation was evaluated by cell counting. Results (1) The DCN extract can significantly inhibit the growth of human hepatoma cells and reduce the mitochondrial metabolic activity; (2) Human hepatoma cells treated with DCN extract for 24 hours, even if replaced with normal medium, their mitochondrial metabolic activity and cells Proliferation was still significantly different from the control. Conclusion The DCN extracts of traditional Chinese medicine containing tumor-suppressing active substances can inhibit the proliferation of cultured hepatoma cells and reduce the mitochondrial metabolic activity.
