目的建立HPLC测定窝儿七中槲皮素、鬼臼毒素、山柰素的方法。方法采用HPLC,Diamonsil(钻石)C18(4.6 mm×250 mm,5μm)色谱柱,甲醇-0.4%的冰醋酸水溶液为流动相,梯度洗脱;检测波长:290 nm,流速:1.0 mL.min-1,柱温30℃。结果槲皮素、鬼臼毒素、山柰素的线性范围分别为0.023~0.287μg(r=0.999 9)、0.208~2.6μg(r=0.999 9)、0.036 1~0.451μg(r=0.999 9),平均回收率分别为99.64%(RSD=0.83%)、100.04%(RSD=0.61%)、99.88%(RSD=1.19%)。结论本实验所建立的方法灵敏、准确、重复性好,可用于窝儿七中槲皮素、鬼臼毒素、山柰素的测定。
OBJECTIVE To establish a HPLC method for the determination of quercetin, podophyllotoxin and kaempferol in nesto seven. Methods HPLC, Diamonsil (C18) column (4.6 mm × 250 mm, 5 μm) and methanol-0.4% glacial acetic acid as mobile phase were used for gradient elution. The detection wavelength was 290 nm and the flow rate was 1.0 mL · min- 1, column temperature 30 ℃. Results The linear ranges of quercetin, podophyllotoxin and kaempferol were 0.023 ~ 0.287μg (r = 0.999 9), 0.208 ~ 2.6μg (r = 0.999 9), 0.036 1 ~ 0.451μg The average recoveries were 99.64% (RSD = 0.83%), 100.04% (RSD = 0.61%) and 99.88% (RSD = 1.19%), respectively. Conclusion The method established in this experiment is sensitive, accurate and reproducible. It can be used for the determination of quercetin, podophyllotoxin and kaempferol in Zhonger seven.
